NimbleGen - Target Enrichment

Enriching target regions of interest for next-generation sequencing

Target enrichment allows focusing of sequencing resources, which leads to reduced cost and simplified analysis. We offer Roche Sample Prep Solutions, from sample collection to library quantification for different sample types and sequencing applications that are proven, complete and simple. Roche Sequencing Solutions offers target enrichment workflows, such as the HyperCap Workflow, and high-quality probes for either of these methods. These workflows enable focusing sequencing resources for targeted resequencing applications in human genetic disease and cancer research.

Product List

Hybridisation-Based

sub_cat_ban_image_amplicon_1880x560_right-copy.pngIn hybridisation or capture-based target enrichment of samples prior to next-generation sequencing (NGS), pools of biotinylated oligonucleotide probes specific for target regions of interest are hybridised to a sequencing library in solution. Next, the biotinylated probe/target hybrids are pulled down by streptavidin-coated magnetic beads to obtain libraries highly enriched for the target regions.

Roche Sequencing Solutions offers the SeqCap® target enrichment products, which use a solution-based capture method for target enrichment. There are target enrichment options for analysing the whole exome or specific disease-associated regions, for investigating methylation changes or for studying transcriptomes.

Benefits

  • A simple, single-step target enrichment solution based on an optimized design algorithm.
  • Various options for the enrichment of the whole exome or regions of  interest, bisulfite-treated DNA or RNA.
  • HyperCap Workflow v2.0 provides an integrated NGS sample preparation solution, combining all reagents and accessories required for library preparation and target enrichment in a single workflow.
  • Predesigned catalog options or custom designs based on specific requirements.
  • An integrated, application-specific sample preparation solution.

Amplicon-Based

sub_cat_ban_image_amplicon_1880x560_right-copy.pngAmplicon-based target enrichment methods for next-generation sequencing (NGS) rely on PCR-amplification of target regions of interest using sequence-specific primers and probes. Using molecular inversion probes (MIP), specific probes consisting of a universal linker flanked by target-specific sequences are constructed. The probes anneal on both sides of the target amplicon and the gap is filled by a DNA polymerase.

Roche Sequencing Solutions offers the HEAT-Seq® portfolio of target enrichment products, which utilise an advanced version of MIP technology, for single nucleotide variation (SNV) detection for targeted sequencing applications in human genetic disease and cancer research.

Benefits

  • Fast and easy workflow with powerful SNV detection.
  • Reliable performance through the use of an advanced version of MIP.
  • Choice between catalog and custom formats  to maximize your time and resources.

HyperCap Workflow

pdp_ban_seqcap_rnachoice_1880x460_right.pngHyperCap products provide an integrated next-generation sequencing (NGS) sample preparation solution that combines all reagents and accessories required for library preparation and target enrichment in a single workflow. The HyperCap Workflow v2.0 integrates high-efficiency KAPA Library Preparation Kits and KAPA Dual-Indexed Adapters with performance-optimised SeqCap® EZ probes and Roche® Universal Blocking Oligo (UBO) Kit. The HyperCap Workflow v2.0 is an improved and streamlined version of our SeqCap EZ HyperCap Workflow, with the convenience of ordering and support from a single vendor, and is automation friendly.

KAPA HyperPrep and KAPA HyperPlus Library Preparation Kits are novel one-tube chemistry with optimally formulated enzymes selected through our directed evolution technology. They enable higher yields of adapter-ligated library and lower amplification bias resulting in higher library diversity, lower duplication rates and more uniform coverage compared to other products available in the market.

Benefits

  • Higher efficiency in library preparation process with lower input amounts.
  • Higher coverage with low duplicate rates.
  • Automation-friendly.
  • Ordering and support of all products of the workflow from a single vendor.
